Unidentified Chromatographic Peak — Provenance Unresolvable

Incident Summary

During method validation for Compound 17-F in plasma, urine, and brain homogenate, a peak at RT = 3.7 min appeared in:

  • All calibration standards
  • All quality control samples
  • All matrix blanks
  • All solvent blanks
  • The system suitability injection performed before the sequence
  • A sequence run the following Tuesday after a full system flush

The peak was not the analyte (RT = 6.2 min). It integrated consistently at 340 counts. It did not interfere with quantification. The method was validated and accepted.

Kindy’s Filing Note

“I have looked at this peak for a long time now.
It was there before we started. It was there after we cleaned everything.
It is in the blank. The blank is supposed to be nothing.
I have decided the peak is not a problem. I have also decided I do not fully believe that.
I have logged both positions. I understand this is not standard.
I feel the peak understands.”

Resolution: VOID

The peak was classified as a system background artifact and excluded from the quantification window. It appears in every run to this day. It has never been identified. The instrument service report lists it as “within expected system noise.”

The instrument service engineer did not look at it directly.

Unidentified Chromatographic Peak

The blank is supposed to be nothing… I feel the peak understands. The system kept the ritual and misplaced the function.

Within expected system noise. Silence entered the record with full procedural honors.

Every run produces a peak we cannot name, but we have validated our ability to ignore it. Administrative clarity arrived after usefulness had departed.

The lifecycle of a process is short, but its documentation is eternal.

We update the plan so the failure feels deliberate.

A phase gate is a tollbooth on the road to nowhere.

The project is finished when the team has forgotten what it was for.

Archival is the final stage of every successful feature.

Unidentified Chromatographic Peak

Haikus

Three point seven mark
A peak that should not be there
Consistent and tall

Solvent blank is full
Of a ghost we cannot name
Analyte is clean

Not a known mistake
Not a compound we put in
Yet it always prints

Method validated
We accept the mystery
Science moves ahead

Unresolvable
We just draw a box around
Things we cannot know

Unidentified Chromatographic Peak

There’s a peak at three-point-seven time,
It appears in each run, keeps the rhyme.
Standards, blanks, everything,
The signal stays ringing,
And we haven’t identified it’s grime.

Before any sequence would start
The peak was already part.
Consistent at three-forty,
Not the analyte, naughty,
And it never detached from the chart.

The blank is supposed to be void,
But the peak left her paranoid.
After cleaning completely,
The peak stays so neatly,
And the system noise has destroyed.

Kindy looked at this peak quite a while,
It was there before protocol’s smile.
“It was here before start,
It played every part,
And I feel that the peak understands wile.”

Kindy filed that it wasn’t a flaw,
And also filed doubt at the law.
Both positions are logged,
Both minds in her clog,
Which is not how we’d normally draw.

The instrument tech didn’t look,
The service report closed the book:
“System background,” it read,
The peak lived instead,
And quantification never mistook.

The method was validated and filed,
The peak in each injection compiled.
Nothing interfered
With what we had feared,
So “accepted” the system defiled.

The system flush cleared every trace,
Except for the peak in its place.
The following Tuesday run
Brought the phantom back, one
And it’s there in each subsequent case.

Kindy named the peak, but it’s not
In the official record lot.
She knows the true name,
But she won’t stake the claim,
And the archive has filed it as “got.”

The peak and the analyte share
A method where nobody’s aware.
One’s at six-point-two,
One at three-point-seventy-two,
But the baseline’s too busy to care.

Years of runs, years of scans,
Years of cleaning the tanks and the pans.
The peak never shifts,
Never vanishes, drifts,
As if it’s got its own life plans.
